Lock-on and -off disulfides provide additional evidence that the aspartate receptor generates a piston-type transmembrane signal. The Cys176-Cys43 and Cys179-Cys39 disulfides lock the signaling helix in the down position and turn the kinase off. The Cys183-Cys39 and Cys197-Cys25 disulfides, by contrast, lock the signaling helix up and trap the kinase in the on state, regardless of ligand occupancy (Chervitz & Falke, 1995, 1996; Falke & Hazelbauer, 2001).
